Description: Neutrophil cytosolic factor 1 (NCF1) is a 47kD cytosolic subunit of the multi-protein complex known as NADPH oxidase. Expression of NCF1 is restricted to granulocytes, monocytes and B lymphocytes. Defects in NCF1 have been reported to be a cause of autosomal recessive chronic granulomatous disease. Applications: Suitable for use in Western Blot, Immunoprecipitation and Immunocytochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:250-1:1000 detected p47-phox in RIPA lysates from HL-60 cells. Immunoprecipitation: 10ul immunoprecipitates p47-phox from 500ug of HL-60 RIPA lysate. Immunocytochemistry: 1:250-1:2000 dilution showed positive cytosolic immunostaining for p47-phox in HL-60 cells fixed with 4% paraformaldehyde and permeabilized with 0.1% Triton X-100. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
