Technical Data
Plexin A3 (SEX)
Plexin A3 (also known as SEX) is a 220 kD member of the Plexin A subfamily of semaphorin receptors. Mature mouse Plexin A3 is predicted to be an 1853 amino acid (aa) type I transmembrane glycoprotein that contains a 1202 aa extracellular domain (ECD) and a 629 aa cytoplasmic region. The ECD contains one semaphorin domain (aa # 34-473), three plexin-semaphorin-integrin (PSI) domains (aa # 492-830), and four lg-like plexin-transcription factor (IPT) repeats (aa # 841-1209). Plexin A3 interacts with neuropilins 1 and 2, and binds class 3 semaphorins. Over aa # 35-150, mouse Plexin A3 is 99% and 97% aa identical to rat and human Plexin A3, respectively.

Western Blot: This antibody can be used at 0.1-0.2ug/ml with the appropriate secondary reagents to detect mouse Plexin A3. The detection limit for rmPlexin A3 is approximately 2 ng/lane under non-reducing and reducing conditions.
Immunohistochemistry: This antibody has been used at a concentration of 10ug/ml to detect Plexin A3 in cryosections of E13 mouse embryos. Tissue was fixed overnight in 4%PFA+14%picric acid in PBS at 4 C. Sections were blocked with 10% donkey serum and 0.1% Triton in 1% BSA in PBS for 45 minutes at room temperature. After blocking, sections were incubated with primary antibody for 3 hours at room temperature followed by NorthernLightsTM 557 donkey anti-goat lgG at room temperature for 1 hour. Between each step, sections were washed with 0.1% BSA in PBS.
Direct ELISA: This antibody can be used at 0.5-1.0ug/ml with the appropriate secondary reagents to detect mouse Plexin A3. The detection limit for rmPlexin A3 is approximately 1.0 ng/well.
Optimal dilutions should be determined by each laboratory for each application.

Storage and Stability:
Lyophilized powder may be stored at 4C for short-term only. Reconstitute to nominal volume by adding sterile PBS with 40-50% glycerol and store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

E. coli-derived recombinant mouse Plexin A3 (aa34-150)
Purified by mouse Plexin A3 affinity chromatography.
Supplied as a lyophilized powder from a 0.2um filtered solution in PBS with 5% trehalose. Reconstitute with sterile PBS, 40-50% glycerol. If 0.5ml is used, the concentration will be 0.2mg/ml.
Selected for its ability to recognize mouse Plexin A3 in the applications listed. In direct ELISAs and Western Blots, this antibody shows less than 5% crossreactivity with rmPlexin A1, rmPlexin A2, rhPlexin B1, rhPlexin C1, and rhPlexin D1.
