Description: SLP 76 was recently identified as a substrate of the T cell receptor stimulated protein tyrosine kinase. This molecule is rapidly phosphorylated on T cell receptor activation. SLP 76 has three identifiable domains; the first containing a number of tyrosines which are the likely sites of phosphorylation, the second a proline-rich domain, and the third an SH2 domain. SLP 76 has been shown to bind to the Grb2 adapter protein in vivo; the association between Grb2 and SLP 76 has been mapped to a 20 amino acid stretch in SLP76's proline-rich region. The SH2 domain of SLP 76 binds to two tyrosine phosphorylated proteins after T cell receptor activation. These molecules are yet to be identified. The SH2 domain of SLP 76 also appears to bind to a serine threonine kinase, perhaps via an indirect association.. Interestingly, overexpression of SLP 76 results in greatly augmented IL-2 production in transiently transfected T cells. Collectively, these data suggest that SLP 76 plays an important role in T cell activation and perhaps is involved in coupling signaling pathways after activation of the T cell antigen receptor. Immunogen: Bacterially expressed GST-SLP 76 fusion protein translated from the cloned human cDNA. Applications: Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilution: Western Blot: 0.5-2ug/ml.. Detects SLP 76 in RIPA lysates from Jurkat cells. Immunoprecipitation: 4ug from a previous lot immunoprecipitated SLP 76 from 500ug of Jurkat cell RIPA lysate. Optimal dilutions to be determined by the researcher. Included Positive Antigen Control: J9000: Jurkat lysate. Add 2.5ul of 2-mercaptoethanol/100ul of lysate and boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane for minigels. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
