Description: Superoxide dismutase (SOD) is an antioxidant enzyme involved in the defense system against reactive oxygen species (ROS). SOD catalyzes the dismutation reaction of superoxide radical anion (O2-) to hydrogen peroxide, which is then catalyzed to innocuous O2 and H2O by glutathione peroxidase and catalase. Several classes of SOD have been identified. These include intracellular copper, zinc SOD (Cu, Zn SOD/SOD1), mitochondrial manganese SOD (Mn SOD/SOD2) and extracellular Cu, Zn SOD (EC SOD/SOD3). The manganese containing 80kD tetrameric enzyme SOD2 is located in the mitochondrial matrix in close proximity to a primary endogenous source of superoxide, the mitochondrial respiratory chain. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications have not been tested. Recommended Dilutions: Western Blot: 0.5-2ug/ml. Detects MnSOD in RIPA lysates of rat L6 RIPA cells. L6 cell lysates were resolved by electrophoresis and transferred to nitrocellulose probed with S8060-10A. Proteins were visualized using an IgG (HRP) Pab Gt xRb and a chemiluminescence detection system. Immunohistochemistry(paraffin): 10ug/ml. Detected MnSOD in paraffin embedded rat brain tissue sections. Optimal dilutions to be determined by the researcher. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
