Syntaxin 5 (STX5, SED5, STX5A) |

Description: STX5 (Syntaxin 5) is a predicted 41-42kD member of the syntaxin family of proteins. It is a t-SNARE that is widely expressed (but not in striated muscle), and embedded in either the cis-Golgi network (a 35kD form) or the ER (the 42kD form). STX5 is involved in the regeneration and maintenance of the Golgi apparatus after mitosis. Human STX5 is a type IV single-pass transmembrane protein (very long cytoplasmic N-terminus) that is 355aa in length. It contains an ER retrieval signal (Arg4Lys5Arg6), a t-SNARE domain (aa263-325) with a coiled-coil region (aa 287-318), and a C-terminal transmembrane sequence. The 35kD short form exhibits an alternate start site at Met55. Two other potential splice variants show either the Met55, or a Met97 start site coupled to a 19aa substitution for aa 303-355. Over aa55-300, human STX5 shares 94% aa identity with mouse STX5. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: Direct ELISA: 0.5-1ug/ml. The detection limit is ~1ng/well. Western Blot: 1ug/ml. Cell lysates were resolved by SDS-PAGE, transferred to an Immobilon-P membrane and immunoblotted. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
