Selective isolation of Vibrio sp. from clinical or food materials.
TCBS is prepared according to the formula of Kobayashi et al. The pH of the medium is elevated to enhance the growth of V. cholerae. Sucrose is fermented by V. cholerae and its biotype Eltor producing yellowing-brown flat colonies. Fishbein et al. reported that V. parahaemolyticus produce light bluish colonies. Enteric bacteria, including Proteus sp. present in fecal specimens are inhibited. TCBS Agar was also found useful in the isolation of V.fluvialis and V. vulnificus. Gram- positive bacteria are inhibited by the bile present in the medium. Bromthymol blue and thymol blue are pH indicators.

Directions: Mix 88 grams of the medium in one Liter of purified water until evenly dispersed. Heat with repeated stirring and boil for one minute to dissolve completely. Poured plates can be prepared after the medium has been cooled to 45-50.0C Do not autoclave. Storage and Stabillty:

Store powdered media at 4C. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept at 4C and used within a short period of time.

Appearance: Supplied as a homogeneous, free flowing and beige to greenish beige powder.

Solubility: clear to trace hazy, forest green solution.

pH: 8.6 at 25.0C
