Description: TIE2 (tyrosine kinase with Ig and EGF homology domains 2) is expressed almost exclusively in endothelial cells in mice, rats and humans. This receptor possesses a unique extracellular domain containing two immunoglobulin-like loops separated by three epidermal growth factor-like repeats that are connected to three fibronectin type III-like repeats. The ligand for the receptor is Angiopoietin 1. Defects in TIE2 are associated with inherited venous malformations; the TIE2 signaling pathway appears to be critical for endothelial cell-smooth muscle cell communication in venous morphogenesis. Applications: Suitable for use in ELISA, Immunoprecipitation, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Immunoprecipitation: Reported to immunoprecipitate Tie2/TEK. Western Blot: 0.2-1ug/ml. HUVEC cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with T5498-72. Proteins were visualized using an IgG (HRP) goat anti-mouse and a chemiluminescence detection system. Immunohistochemistry: Reported to detect Tie2/TEK in frozen, Triton-treated sections. Not suitable for use in paraf n- embedded sections. Optimal dilutions to be determined by the researcher. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
