Transforming Growth Factor beta1 (TGF beta 1, TGFb1, Camurati Engelmann Disease, CED, Diaphyseal Dysplasia 1 Progressive, DPD1, TGFb1, Differentiation inhibiting factor, Cartilage-inducing factor)
TGF-b1, -2 and -3 are a closely related group of proteins (70-80% sequence homology) that are produced by many cell types and function as growth and differentiation factors. The active forms of TGF-b1, -2 and -3 are disulfide-linked homodimers.

Suitable for use in Capture ELISA, Neutralization, Western Blot, Flow Cytometry and Immunohistochemistry. Other applications not tested.

Recommended Dilutions:
Capture ELISA: 2-8ug/ml. Can be paired with T8250-16B (0.1-0.4ug/ml) as the detection antibody in sandwich ELISA.
Neutralization: 0.3-1ug/ml will neutralize 50% of the bioactivity in the presence of 0.25ng/ml of recombinant human TGFb1 and 7.5ng/ml recombinant mouse IL-4. Measured by its ability to neutralize TGF-b1 inhibition of IL-4 dependent proliferation in the HT-2 mouse T cell line.
Western Blot: 1ug/ml. Reducing conditions are not recommended.
Flow Cytometry (Intracellular): 2.5ug/ml labels 1x10e6 cells. Validated using PC-3 human prostate cancer cell line fixed with paraformaldehyde and permeablized with saporin.
Immunohistochemistry: 8-25ug/ml in immersion fixed paraffin embedded sections of human prostate cancer tissue.
Optimal dilutions to be determined by the researcher.

Storage and Stability:
Lyophilized powder may be stored at -20C. Stable for 12 months after receipt at -20C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Purified by Protein G affinity chromatography. Endotoxin: <0.1EU/ug (LAL)
Supplied as a lyophilized powder from PBS, pH 7.4, 5% trehalose. Reconstitute with 1ml sterile PBS.
Recognizes human TGF-b1 both in its active form and in its latent form when it is complexed with LAP (latency associated peptide.). Neutralizes the biological activity of TGF-b1 and TGF-b1.2. Species Crossreactivity: mouse and rat. In sandwich ELISA, <2% crossreactivity with recombinant human TGF-b3 and recombinant amphibian TGF-b5 was observed and no crossreactivity with recombinant porcine or recombinant human TGFb2 was observed.
