Dextrose Tryptone Broth is a non-selective medium which can support the growth of a variety of mesophilic and thermophilic microorganisms. It contains the pH indicator bromcresol purple. Inoculated filter membranes are incubated on this medium at 55°C to detect and enumerate thermophilic "flat sour" sporulating microorganisms. Isolation and enumeration of “flat-sour” thermophiles and mesophiles in food by membrane filter technique.
Suspend 30g of the powder in 1L of distilled water. Mix thoroughly. Heat with frequent agitation and boil for 1 minute to completely dissolve the powder. Dispense into appropriate containers, loosen caps and autoclave for 15 minutes at 121ºC (15psi).
Store powdered media at 4ºC. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept at 4°C and used within a short period of time.
