A Proliferation Inducing Ligand, TNFSF13, CD256, TALL-2

APRIL is a cytokine that belongs to the TNF superfamily and binds to TACI and BCMA. It is implicated in the regulation of tumor cell growth and is involved in monocyte/macrophage-mediated immunological processes. APRIL is an antibody developed by antibody phage display technology using a human naive antibody gene library. These libraries consist of scFv (single chain fragment variable) composed of VH (variable domain of the human immunoglobulin heavy chain) and VL (variable domain of the human immunoglobulin light chain) connected by a polypeptide linker. The antibody fragments are displayed on the surface of filamentous bacteriophage (M13). This scFv was selected by affinity selection on antigen in a process termed panning. Multiple rounds of panning are performed to enrich for antigen-specific scFv-phage. Monoclonal antibodies are subsequently identified by screening after each round of selection. The selected monoclonal scFv is cloned into an appropriate vector containing a Fc portion of interest and then produced in mammalian cells to generate an IgG like scFv-Fc fusion protein.

Applications
Suitable for use in ELISA, Neutralization and Immunoprecipitation. Other applications not tested.
Recommended Dilution
Immunoprecipitation: 1:200 Neutralization: Inhibits the binding of mouse APRIL to BCMA and TACI. Optimal dilutions to be determined by researcher.
Endotoxin
<0.01EU/ug (LAL-test; Lonza)
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Mouse APRIL (aa98-232)
Form
Supplied as a liquid in PBS, 10% glycerol. No preservative added.
Purity
Purified by Protein A affinity chromatography from HEK293 cell culture supernatant.
Specificity
Recognizes mouse APRIL. Does not recognize mouse BAFF.
