The caspases are a group of cysteine enzymes, which cleave proteins in response to intrinsic and extrinsic pathways that cause apoptotic cell death. The caspases can be grouped into two subgroups based on their roles in apoptosis. Initiator caspases (caspases 2, 8, 9, and 10) are activated through the apoptosis-signaling pathways and activate the effector caspases (caspases 3, 6, and 7) which carry out apoptosis. Caspase cascades are initiated through assembly of multiprotein complexes that trigger activation of the initiator caspases, which are then released and are able to activate the downstream effector caspases.

Applications
Suitable for use in Western Blot and Immunocytochemistry. Other applications not tested.
Recommended Dilution
Western Blot: 1:1000 Immunocytochemistry: 1:100 Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Recombinant protein corresponding to amino acid residues in the N-terminal region of human Caspase-3.
Form
Supplied as liquid in PBS, 1mg/ml BSA, 0.05% sodium azide, 50% glycerol.
Purity
Purified by Protein A affinity chromatography.
Specificity
Recognizes human Caspase 3. Detects a 32kD protein corresponding to the molecular weight of Caspase-3 on SDS-PAGE immunoblots of human Jurkat and K562 cells.
