Cytokine that plays an essential role in the regulation of survival, proliferation and differentiation of hematopoietic precursor cells, especially mononuclear phagocytes, such as macrophages and monocytes. Promotes the release of proinflammatory chemokines, and thereby plays an important role in innate immunity and in inflammatory processes. Plays an important role in the regulation of osteoclast proliferation and differentiation, the regulation of bone resorption, and is required for normal bone development. Required for normal male and female fertility. Promotes reorganization of the actin cytoskeleton, regulates formation of membrane ruffles, cell adhesion and cell migration. Plays a role in lipoprotein clearance.
Applications
Suitable for use in ELISA, Western Blot and Neutralization. Other applications have not been tested.
Recommended Dilution
Neutralization: <0.1ug/ml will yield one-half maximal inhibition [ND50] of the biological activity of human CSF1 (1ng/ml). ELISA (Sandwich): 0.5-2ug/ml using 100ul/well antibody solution in conjunction with biotinylated anti-human CSF1 as a detection antibody, allows the detection of at least 0.2-0.4ng/well of recombinant human CSF1 Western Blot: 0.1-0.2ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant human CSF1 is 1.5-3ng/lane, under either reducing or non-reducing conditions. Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Recombinant protein corresponding to human CSF1 expressed in E. coli.
Form
Supplied as a lyophilized powder from PBS, pH 7.2. Reconstitute with sterile ddH2O to a concentration of 0.1-1mg/ml.
Purity
Purified by Immunoaffinity chromatography. |Endotoxin: <0.1ng/ug protein (<1EU/ug)
Specificity
Recognizes human CSF1.
