GCLC, Glutamate--cysteine ligase catalytic subunit, is an enzyme that in humans is encoded by the GCLC gene.Glutamate-cysteine ligase, also known as gamma-glutamylcysteine synthetase is the first rate limiting enzyme of glutathione synthesis. The enzyme consists of two subunits, a heavy catalytic subunit and a light regulatory subunit. The gene encoding the catalytic subunit encodes a protein of 367 amino acids with a calculated molecular weight of 72.773 kDa and maps to chromosome 6p12.1. Deficiency of gamma-glutamylcysteine synthetase in human is associated with enzymopathic hemolytic anemia.

Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested.
Recommended Dilution
Immunohistochemistry: paraffin Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
A synthetic peptide corresponding to a sequence at the N-terminal of human GCS/GCLC, different from the related rat and mouse sequences by one amino acid.
Form
Supplied as a lyophilized powder from PBS, 5% BSA, 0.05% Thimerosal, 0.05% sodium azide. Reconstitutie with 200ul sterile ddH2O.

Purified by immunoaffinity chromatography.
Specificity
Recognizes human, mouse and rat GCS/GCLC. No crossreactivity with other proteins.
