CAPN1 is also known as CANP or muCL. The calpains, calcium-activated neutral proteases, are nonlysosomal, intracellular cysteine proteases. The mammalian calpains include ubiquitous, stomach-specific, and muscle-specific proteins. The ubiquitous enzymes consist of heterodimers with distinct large, catalytic subunits associated with a common small, regulatory subunit. This gene encodes the large subunit of the ubiquitous enzyme, calpain 1. Several transcript variants encoding two different isoforms have been found for this gene.

Applications
Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested.
Recommended Dilutions
Western Blot: 0.1-0.5ug/ml, the detection limit is ~0.1ng/lane under reducing conditions. Immunohistochemistry (Paraffin): 0.5-1ug/ml, boiling the paraffin sections in 10mM citrate buffer, pH 6.0, for 20 minutes is required for the staining of formalin/paraffin sections. Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile dH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Recombinant protein corresponding to human Calpain 1 expressed in E. coli. (Position: Q396-A555). Species Sequence Homology: mouse and rat, 86%
Form
Supplied as a lyophilized powder from PBS 5% BSA, 0.05% sodium azide. Reconstitute with 200ul sterile ddH2O.
Purity
Purified by immunoaffinity chromatography.
Specificity
Recognizes human Calpain 1. Species Crossreactivity: mouse and rat
