SHC, also known as SHC1 (SHC-transforming protein 1) or SHCA, is a protein that in humans is encoded by the SHC1 gene. SCOP classifies the 3D structure as belonging to the SH2 domain family. This gene encodes three main isoforms that differ in activities and subcellular location. While all three are adapter proteins in signal transduction pathways, the longest (p66Shc) may be involved in regulating life span and the effects of reactive oxygen species. The other two isoforms, p52Shc and p46Shc, link activated receptor tyrosine kinases to the Ras pathway by recruitment of the GRB2/SOS complex. p66Shc is not involved in Ras activation. Unlike the other two isoforms, p46Shc is targeted to the mitochondrial matrix. Several transcript variants encoding different isoforms have been found for this gene.

Applications
Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested.
Recommended Dilution
Western Blot: 0.1-0.5ug/ml, the detection limit is ~0.1ng/lane under reducing conditions. Immunohistochemistry (frozen): 0.5-1ug/ml Immunohistochemistry (paraffin): 0.5-1ug/ml, boiling the paraffin sections in 10mM citrate buffer, pH 6.0, for 20 mins is required for the staining of formalin/paraffin sections. Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Synthetic peptide corresponding to aa536-564, VDPEGVVRTKDHRFESVSHLISYHMDNHL, from human SHC at C-terminus, identical to the related mouse and rat sequences.

Supplied as a lyophilized powder from PBS, 5% BSA, 0.05% sodium azide. Reconstitute with 200ul sterile dH2O.

Purified by immunoaffinity chromatography.
