GCLC, also named Glutamate--cysteine ligase catalytic subunit, is an enzyme that in humans is encoded by the GCLC gene. Glutamate-cysteine ligase, also known as gamma-glutamylcysteine synthetase is the first rate limiting enzyme of glutathione synthesis. The enzyme consists of two subunits, a heavy catalytic subunit and a light regulatory subunit. The gene encoding the catalytic subunit encodes a protein of 367aa with a calculated molecular weight of 72.773kD and maps to chromosome 6p12.1. Deficiency of gamma-glutamylcysteine synthetase in human is associated with enzymopathic hemolytic anemia.

Applications
Suitable for use in Western Blot, Immunohistochemistry. Other applications not tested.
Recommended Dilution
Western Blot: 0.1-0.5ug/ml Immunohistochemistry (paraffin): 0.5-1ug/ml Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recombinant protein corresponding to human GCLC expressed in E. coli. Species sequence homology: mouse and rat (94%)
