Adipophilin is a ubiquitous compound of lipid droplets. Adipophilin is present in milk fat globule membranes and on the surface of lipid droplets in various cultured cell lines and is inducible by etomoxir. Enhanced expression of ADRP is a useful marker for pathologies characterized by increased lipid droplet accumulation; such diseases include atheroma, steatosis, obesity and certain cases of liposarcoma. It can also be a potent marker for atherosclerosis. It can also be used to study the virus entry of e.g. HCV via lipid droplets. Adipophilin is localized to glandular cells of lactating mammary gland (ductal cells are negative), zona fasciculata of the adrenal gland, Sertoli cells of the testis, and in fat-accumulating hepatocytes of alcoholic cirrhotic fatty liver. Adipocytes do not display staining. Lipid-storing CD68-positive macrophages do positively stain for adipophilin.

Applications
Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested.
Recommended Dilutions
Immunohistochemistry (Frozen tissue; See reference 14 and paraffin-embedded tissue after microwave treatment): 1:10 in PBS, pH 7.4. Incubate for 1 hour at RT or overnight at 4°C. Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Synthetic peptide corresponding to aa5-27 from the N-terminus of human Adipophilin. MW: 48.1kD. Apparent MW: 52kD by SDS-PAGE.
Form
Supplied as a lyophilized powder from PBS, pH 7.4, 0.5% BSA, 0.09% sodium azide. Reconstitute with 1ml sterile ddH2O.
Purity
Purified by Protein A affinity chromatography.
Specificity
Recognizes human Adipophilin. Reactive with Caco, PLC, HaCat, SV80, RD125, Huvec, RV, PC-12 (rat adrenal gland) and MDCK cultured cell lines. Negative with glioma. Species Crossreactivity: rat and canine. Does not crossreact with bovine or mouse.
