Arfaptin 2, or ADP-ribosylation factor interacting protein 2, is a Rac1 binding protein necessary for Rac-mediated actin polymerization and the subsequent formation of membrane ruffles and lamellipodia.1 Arfaptin-2 has been shown to be involved in interactions between the Rac and ARF signaling pathways.2 Arfaptin 2 also regulates the aggregation of mutant Huntingtin protein by possibly impairing proteasome function. Expression of Arfaptin 2 was shown to be increased at sites of neurodegeneration. Mutant Huntingtin protein is characteristic of Huntington’s disease, an inherited neurodegenerative disorder.3
Applications
Suitable for use in Immunofluorescence and Western Blot. Other applications not tested.
Recommended Dilution
Immunofluorescence: 0.3-7ug/ml Western Blot: 1-3ug/ml Optimal dilutions to be determined by the researcher.
Positive Control: human HeLa cervical carcinoma, A549 lung carcinoma, MCF-7 breast adenocarcinoma, T47D breast ductal carcinoma, NCI-H69 lung carcinoma, rat PC12, and GFP-Arfaptin 2-transfected HeLa cell lysates.
Storage and Stability
May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add glycerol (40-50%), aliquot and store at -20°C or colder. Aliquots are stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Note: Applications are based on unconjugated antibody.
