Human CD16 is a low affinity receptor for complexed IgG and occurs in two distinct forms, as a glycosyl phosphtidyl inositol (GPI) anchored protein in granulocytes, and as an integral membrane protein on NK cells and macrophages.

Applications
Suitable for use in Flow Cytometry. Other applications not tested.
Recommended Dilution
Optimal dilutions to be determined by the researcher.
Protocol
1. Five x 10e5 ficoll prepared human peripheral blood lymphocytes were washed and pre incubated 45 minutes with 80ul of C2267-33A at 1ug/ml. Cells were washed twice and incubated with  IgG (FITC) Goat anti-Mouse, after which they were washed three times, fixed and analyzed by FACS. A net 14% sub population of the cells stained positive with a mean shift of 1.8 log10 fluorescent units when compared to a Mouse IgG1 negative control at a similar concentration.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Human polymorphonuclear leukocytes.
Form
Supplied as a liquid in 50mM sodium phosphate pH 7.5, 100mM potassium chloride, 150mM sodium chloride, 0.5mg/ml gentamicin sulfate.
Purity
Purified by Protein A affinity chromatography from tissue culture supernatant.
Specificity
Recognizes both allelic forms of CD16. Antibody blocks binding of complexed IgG to CD16.
