The CD22 antigen is only expressed on the cell surface of mature B-lymphocytes. The CD22 antigen is expressed on >90% of peripheral blood B-cells (1). CD22 is expressed on most peripheral B-cell lymphomas and leukemias including hairy cell leukemia, but not on the immature bone marrow type of leukemia. This antigen is not present on T-cell, monocytes or macrophages.
Applications
Suitable for use in ELISA and Western Blot. Other applications have not been tested.
Recommended Dilutions
ELISA (Sandwich): 0.5-2ug/ml using 100ul/well antibody solution) In conjunction with biotinylated anti-human CD22 (C2272-98G) as a detection antibody, allows the detection of at least 0.2-0.4ng/well of recombinant human CD22. Western Blot: 0.1-0.2ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant human CD22 is 1.5-3ng/lane, under either reducing or non-reducing conditions. Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Recombinant protein corresponding to human CD22 expressed in CHO cells.
Form
Supplied as a lyophilized powder from PBS, pH 7.2. Reconstitute with sterile ddH2O to a concentration of 0.1-1mg/ml.
Purity
Purified by immunoaffinity chromatography. Endotoxin: ≤1EU/ug
Specificity
Recognizes human CD22.
