Recognizes CD68 which is important for identifying macrophages in tissue sections. It stains macrophages in a wide variety of human tissues, including Kupffer cells and macrophages in the red pulp of the spleen, in lamina propria of the gut, in lung alveoli, and in bone marrow. Reacts with myeloid precursors, peripheral blood granulocytes and plasmacytoid T cells which are supposed to be of monocyte/macrophage origin. Shows strong granular cytoplasmic staining of chronic and acute myeloid leukemia and also reacts with rare cases of true histiocytic neoplasia. Tumors of lymphoid origin are usually not stained.

Applications
Suitable for use in Immunohistochemistry and Immunofluorescence. Not recommended for use in Western Blot. Other applications not tested.
Recommended Dilution
Immunohistochemistry (Paraffin): 1:40-1:80. Requires antigen retrieval using heat treatment prior to staining of paraffin sections; 1mM EDTA, pH 8.0, is recommended. Immunohistochemistry: Frozen sections Optimal dilutions to be determined by the researcher.
Hybridoma
p3-NS1-Ag4-1 myeloma cells with spleen cells from Balb/c mice.
Positive Control
Human Tonsil
Recommended Secondary Antibodies
I1904-65W: IgG, F(ab’)2 (HRP) Pab Rb x Mo I1904-66C: IgG, F(ab’)2, X-Adsorbed (HRP) Pab Rb x Mo
Recommended Negative Control
I1904-84Q3 IgG2b Murine Negative Control
Storage and Stability
Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Fusion protein corresponding to external domain of human CD68.
Form
Supplied as a lyophilized powder, 0.09% sodium azide. Reconstitute with 1ml sterile ddH2O.
Purity
Supernatant
Specificity
Recognizes the human CD68 cell surface antigen, a 110kD glycoprotein primarily expressed by macrophages and monocytes.
