MaxLight™650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor™647, DyLight™649, Cy5™ and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (655nm); Emission (676nm); Extinction Coefficient 250,000.
Expression of cdc2 plays a critical role in cell transition through the G2/M phase. The activity of cdc2 in dividing cells increases at the onset of M phase coincident with disassembly of the cell nucleus, generation of mitotic spindles and chromosome condensation. Several proteins including Histone H1 and pp60 src are substrates for cdc2.
Applications
Suitable for use in Western Blot, Immunoprecipitation and Immunocytochemistry. Other applications not tested.
Recommended Dilution
Western Blot: 0.5-2ug/ml detected cdc2 from human A431, mouse 3T3 and rat L6 RIPA cell lysates. Immunoprecipitation: 4ug Immunoprecipitates cdc2 from 500ug of A431 RIPA lysates. Optimal dilutions to be determined by the researcher.
Included Positive Antigen Control
A0001-50: Non-stimulated A431 lysate. Use 20ug per lane for minigels
Storage and Stability
Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™650 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
Immunogen
Synthetic peptide corresponding to aa 42-57, EGVPSTAIREISLLKE-GGC, of human cdc2 kinase. The immunogen is identical in a wide range of species.
Form
Supplied as a liquid in PBS, pH 7.2. No preservative added. Labeled with MaxLight™650.
Purity
Purified by antigen affinity chromatography.
Specificity
Recognizes human cdk1/cdc2 kinase (34kD). Species crossreactivity: mouse and rat. Species sequence homology: monkey, drosophilia, canine, sheep, chicken, frog and starfish. Wide species crossreactivity is expected.
