HS1, also called hematopoietic cell-specific LYN substrate 1 (HCLS1), LckBP1, p75, and hematopoietic lineage cell-specific protein, is expressed only in tissues and cells of hematopoietic origin (1,2). HS1 contains 4 cortactin repeats and one SH3 domain (2). It is phosphorylated after activation of immune receptors (3-5), leading to its recruitment to the immune synapse. Phosphorylation of HS1 is also required for regulating actin dynamics and provides docking sites for many other signaling molecules, such as Vav1 and PLCg1 (6). HS1 also plays an important role in platelet activation (7).

Applications
Suitable for use in Flow Cytometry, Western Blot and Immunoprecipitation. Other applications not tested.
Recommended Dilution
Western Blot: 1:1000 Immunoprecipitation: 1:50 Flow Cytometry: 1:25 Optimal dilutions to be determined by the researcher.
Storage and Stability
For long-term storage, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Synthetic peptide corresponding to residues surrounding Pro310 of human HS1.
Form
Supplied as a liquid in 10mM sodium HEPES, pH 7.5, 150mM sodium chloride, 0.1mg/ml BSA, 50% glycerol.
Purity
Purified by peptide affinity chromatography.
Specificity
Detects endogenous levels of total HS1 protein. This antibody does not recognize mouse and rat HS1 proteins. HS1 has a calculated size of 54kD, but has an apparent molecular weight of 80kD on SDS-PAGE gels.
