MaxLight™650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor™647, DyLight™649, Cy5™ and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (655nm); Emission (676nm); Extinction Coefficient 250,000.

Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the 'beads on a string' structure. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine. Acetylation of histone H3 occurs at several different lysine positions in the histone tail and is performed by a family of enzymes known as Histone Acetyl Transferases (HATs). Acetylation of lysine14 is commonly seen in genes that are being actively transcribed into RNA.
Applications
Suitable for use in Western Blot, Immunocytochemistry and Chromatin Immunoprecipitation. Other applications not tested.
Recommended Dilutions
Western Blot: 0.05ug/ml detected Histone H3 on 10ug of HeLa acid extract, sodium butyrate treated HeLa cells and recombinant Histone H3. Proteins were visualized using an IgG (HRP) goat anti-rabbit and a chemilumnescence detection system. Immunocytochemistry: 1:500 detected acetylated Histone H3 in NIH/3T3 and A431 cells. Chromatin Immunoprecipitation: 5ug using HeLa extracted chromain Optimal dilutions to be determined by the researcher.

Storage and Stability
Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™650 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.

Synthetic peptide corresponding to aa1-20 of Tetrahymena histone H3 (ARTKQTAR[K*]STGG[K*]APRKQLC: where K* is acetylated) (KLH).

Specificity
