Interleukin 17 (cytotoxic T-lymphocyte-associated serine esterase 8) is a proinflammatory cytokine produced by activated T cells. This cytokine regulates the activities of NF-kappaB and mitogen-activated protein kinases. This cytokine can stimulate the expression of IL6 and cyclooxygenase-2 (PTGS2/COX-2), as well as enhance the production of nitric oxide (NO). High levels of this cytokine are associated with several chronic inflammatory diseases including rheumatoid arthritis, psoriasis and multiple sclerosis. Interleukin-17 (IL-17) and its cognate receptor, IL- 17R, are examples of cytokine receptor pairs. IL-17 protein exhibits a striking degree of homology with the HSV13 protein which mimics its function.

Applications
Suitable for use in ELISA, Western Blot and Functional Assays. Other applications not tested.
Recommended Dilution
ELISA (sandwich): 0.5-2ug/ml, ussing 100ul/well antibody solution) a concentration of 0.5-2ug/ml of this antibody is required. In conjunction with I8439-04R as a detection antibody, allows the detection of at least 0.2-0.4 ng/well of recombinant hIL-17A. Western Blot: 0.1-0.2ug/ml, used in conjunction with compatible secondary reagents the detection limit for recombinant hIL-17A is 1.5- 3.0 ng/lane, under either reducing or non-reducing conditions. Optimal working dilutions to be determined by researcher.
Matched Pair
Detection: I8439-04R: Interleukin 17 (Biotin) Capture: I8439-07: Interleukin 17
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Recombinant protein corresponding to human IL-17, expressed in E. coli.
Form
Supplied as a lyophilized powder from PBS, pH 7.4. Reconstitute with 100ul sterile ddH2O. For long term storage the addition of 0.09% sodium azide is recommended.
Purity
Purified by immunoaffinity chromatography.
Specificity
Recognizes human interleukin-17.
