Keratin 10 (Cytokeratin 10, CK 10) is expressed in all suprabasal layers of the epidermis. In the epidermis, expression of CK 10 strictly parallels the extent of differentiation; it is absent in the basal layer, appears in the first suprabasal layers and increases in concentration towards the granular layer. Expression of CK 10 is related to maturation of malignant keratinocytes, being preferentially detected in more differentiated parts.

Applications
Suitable for use in Immunofluorescence, Western Blot, and Immunohistochemistry. Other applications have not been tested.
Recommended Dilutions
Western Blot: 1-2ug/ml for 2 hours at RT Immunohistochemistry (formalin/paraffin): 2-4ug/ml for 30 minutes at RT. No special pretreatment is required for staining of formalin-fixed, paraffin-embedded tissues Optimal dilutions to be determined by the researcher.

Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Cytoskeletal preparation extracted from human ectocervical epithelium. Cellular Localization: Cytoplasmic.
Form
Supplied as a liquid in 10mM PBS, pH 7.4, 0.2% BSA, 15mM sodium azide. Also available with BSA and azide. See K0199-12.
Purity
Purified by Protein G affinity chromatography.
Specificity
Recognizes human Keratin 10 at 56.5kD. Species Crossreactivity: rat, canine and feline.
