Lat is an adapter protein expressed in lymphoid cells, which is a substrate for ZAP70 and Syk protein kinases. Phosphorylated LAT associates with GRB2, PLC-gamma1 and P85 PI 3-K. Expression of a mutant LAT protein missing tyrosine phosphorylation sites leads to inhibition of T-cell activation, suggesting that the protein is an important signal transducer in T-cells.

Applications
Suitable for use in Western Blot and Immunoprecipitation. Other appllications not tested.
Recommended Dilutions
Western Blot: 2-8ug/ml detects LAT in RIPA lysates from Jurkat cells. Add 2.5ul of 2-mercaptoethanol/100ul of lysate and boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane for minigels. Jurkat cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with anti-LAT (1mg/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. Immunoprecipitation: 2ug immunoprecipitates LAT from 500ug of Jurkat RIPA lysate. Optimal dilutions to be determined by the researcher.
Positive Antigen Control
Jurkat cell lysate.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
GST fusion protein corresponding to residues 31-233 of human LAT
Form
Supplied as a liquid in 0.1M Tris-glycine, pH 7.4, 0.15M sodium chloride; 0.05% sodium azide.
Purity
Purified by Protein G affinity chromatography.
Specificity
Recognizes both forms of human LAT, Mr 36/38kD.
