MCP-1 is a 99 amino acid, including a 23aa signal peptide, ~13kD chemotactic protein, a chemoattractant for monocytes and basophils but not neutrophils or eosinophils (Weber et al.1996).

MCP-1 is produced by a number of cell types, including monocytes, macrophages, microglia, fibroblasts and epithelial cells (Cushing et al. 1990). MCP-1 influences T-cell immunity polarizing responses towards the Th2 phenotype (Karpus and Kennedy 1997). Variations in the CCL2 gene affects susceptibility to Mycobacteriun tuberculosis infection (Flores-Villanueva et al. 2005).
Applications
Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested.
Recommended Dilution
ELISA: 1-2ug/ml Immunohistochemistry (Paraffin): 10ug/ml. Requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH 6.0 is recommended for this purpose. Western Blot: 0.2-0.4ug/ml Optimal dilutions to be determined by the researcher.
Immunohistochemistry Positive Control Tissue
Human Breast Invasive Ductal Carcinoma
Hybridoma
Spleen cells from immunized BALB/c X ICR F1 mice were fused with cells of a mouse myeloma cell line.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Recombinant protein corresponding to human MCP-1.
Form
Supplied as a lyophilized powder in PBS. No preservative added. Reconstitute with 500ul sterile ddH2O.
Purity
Purified by Protein A affinity chromatography from tissue culture supernatant.
Specificity
Recognizes human MCP-1. Does not cross-react with MCP-2, MCP-3 or other structurally related chemokines.
