Mitogen Activated Protein Kinase Kinase 5, EC=2.7.12.2
MEK5 (also known as MKK5) is a dual specificity serine/threonine protein kinase belonging to the MAP kinase kinase family. MEK5 has been shown to specifically activate ERK5 (Zhou et al., 1995) whereas MEK5 itself is regulated by MEKK3 (Chao et al., 1999). An important link between MEK5 and metastatic prostrate cancer has been demonstrated (Mehta et.al., 2003). Dual phosphorylation of Ser-311 and Thr-315 have been implicated in cell proliferation (Cameron et al., 2004).
Applications
Suitable for use in Western Blot. Other applications not tested.
Recommended Dilution
Western Blot: 1:1000 detects a band of ~49kD in rat testes cell lysates. Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Synthetic phospho-peptide corresponding to amino acid residues surrounding Ser311/Thr315 of rat MEK5, conjugated to KLH. Species sequence homology: Mouse, canine, Primate, Xenopus, Zebrafish, Human, Bovine, Chicken

Supplied as a liquid in 10mM HEPES, pH 7.5, 150mM sodium chloride, 0.1mg/ml BSA, 50% glycerol.
Purity
Purified by affinity chromatography.
Specificity
Recognizes endogenous levels of the ~49kD rat MEK5 protein phosphorylated at Ser311, Thr315. Immunolabeling is blocked by preadsorption with the phosphopeptide used as antigen, but not by the corresponding non-phosphopeptide.
