The NMDA receptor (NMDAR) is an ionotropic receptor for glutamate (NMDA (N-methyl D-aspartate) is a name of its selective specific agonist). Activation of NMDA receptors results in the opening of an ion channel that is nonselective to cations. This allows flow of Na+ and small amounts of Ca2+ ions into the cell and K+ out of the cell. Calcium flux through NMDARs is thought to play a critical role in synaptic plasticity, a cellular mechanism for learning and memory. The NMDA receptor is distinct in that it is both ligand-gated and voltage-dependent.

Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested.

Western Blot: 0.5-2ug/ml detects NR2A (~170kD) in rat brain microsomal preparations. Rat brain microsomal preparation was resolved by electrophoresis, transferred to nitrocellulose and probed with N5380-25. Proteins were visualized using an IgG (HRP) goat anti-rabbit and a chemiluminescence detection system. Immunoprecipitation: 4ug immunoprecipitates NR2A from rat brain hippocampus membranes and rat brain microsomal preparations. Optimal dilutions to be determined by the researcher.

Brain Tissue

Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™490 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.

Note: Applications are based on unconjugated antibody.
