PCOLE1 (PCPE-1) is a secreted 55kD enzyme adaptor that enhances the activity of the BMP-1/tolloid family of metalloproteinases (also known as procollagen C-proteinases) on fibrillar procollagens. PCOLE1 binds to the C-propeptide region and also elsewhere in the procollagen molecule to specifically stimulate BMP-1/tolloid processing of procollagen I, II, and III.
Applications
Suitable for use in ELISA, Western Blot and Immunoprecipitation. Other applications not tested.
Recommended Dilutions
Western Blot: 0.1ug/ml Immunoprecipitation: 25ug/ml using conditioned cell culture medium spiked with Recombinant Mouse PCPE‑1 Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Recombinant protein corresponding to Gln25-Ala468 (predicted) from mouse PCOLE1, expressed in NS0 cells.
Form
Supplied as a lyophilized powder from PBS, pH 7.4, 5% trehalose. Reconstitute with 500ul sterile PBS.
Purity
Purified by immunoaffinity chromatography.
Specificity
Recognizes mouse PCOLE1. Shows ~35% crossreactivity with recombinant human PCPE.
