Pepsinogen I, a zymogen, or proenzyme secreted by Chief cells in the stomach, is cleaved to form pepsin both in an autocatalytic fashion and by pepsin itself. In humans there are two related forms of pepsin, Pepsinogen I, also known as Pepsinogen A, and Pepsinogen II, also known as Pepsinogen B or progastricsin.

A protein precursor to pepsin, Pepsinogen I or Pepsinogen A is secreted exclusively by the chief cells of the gastric corpus. The levels of Pepsinogen I correlate directly with the active number of chief cells in this area. In cases of severe atrophic gastritis of the corpus region, the chief cells will be destroyed and the levels of Pepsinogen I will fall dramatically.
Applications
Suitable for use in FLISA, Immunohistochemistry and RIA. Other applications have not been tested.
Recommended Dilutions
Immunohistochemistry: Paraffin sections. Antigen retrieval using heat treatment prior to stainin of paraffin sections. Sodium citrate pH 6.0 is recommended. Optimal dilutions to be determined by the researcher.
Affinity Constant
4x10e10L/mol
Recommended Secondary Antibodies
I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo I1904-06H IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo
Storage and Stability
Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: APC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
Immunogen
Human pepsinogen I (native).
Form
Supplied as a liquid in PBS, pH 7.2. Labeled with Allophycocyanin (APC).
Purity
Purified by Protein A affinity chromatography from tissue culture supernatant
Specificity
Recognizes human pepsinogen I, both zymogen and proenzyme.
