PLC-d3 (phospholipase C-delta 3) is an 105kD member of the d-type PLC isozyme family. It is expressed in corneal epithelium, platelets, cardiac and skeletal muscle. PLC-d3 preferentially hydrolyzes membrane PIP release, and DAG, activating PKC. Human PLC-d3 is 789aa in length. It contains a plextrin homology domain that tethers the enzyme to the membrane (aa63-172), three EF-hand motifs that spatially orient the molecule (aa182-285), a catalytic region (aa337-644), and a C2 domain that secures the catalytic region to its substrate (aa647-752). There are multiple potential isoforms. There is an alternate start site at Met54, a 40aa substitution for the C-terminal 248 aa, and a Leu substitution for aa102-109. Over aa148-344, human PLC-d3 is 93% aa identical to mouse PLC-d3.
