PAPP-A was initially identified as a high molecular mass component in human pregnancy serum. In pregnancy serum PAPP-A exists in a disulfide bound heterotetrameric complex consisting of two 200kD PAPP-A subunits and two 50-90kD subunits of the proform of eosinophil major basic protein (proMBP). The concentration of PAPP-A in serum increases with gestational age, however, in Down’s syndrome pregnancies PAPP-A concentration is markedly reduced in the first trimester. As such, PAPP-A is widely used as an early marker for Down’s syndrome. PAPP-A has also been shown to exist in a homodimeric, 400kD form in unstable atherosclerotic plaques. Increased circulating concentrations of this dimeric PAPP-A are associated with acute coronary syndromes.

Hybridoma
Derived from hybridization of SP2/0 myeloma cells with spleen cells of Balb/c mice immunized with purified human PAPP-A/proMBP complex.
Storage and Stability
Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: PE conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
