Protein C is a vitamin K-dependent serine protease produced in the liver and made up of 2 polypeptide chains. The 62kD proenzyme is activated by thrombin bound to an endothelial surface receptor and the active enzyme cleaves factor Va and VIIIa and thus inhibits blood coagulation. The molecular weight of the active enzyme is 55kD and the normal concentrations in human plasma is approximately 1-3ng/ml because of the very fast turnover, the proenzyme concentration is ~3ug/ml.

Applications
Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested.
Recommended Dilutions
ELISA: Strong reaction is seen in capture ELISA in combination with a biotin labeled antigen against Protein C only when using a tris-buffer containing 2.5mM CaCl2 Western Blot: After SDS-PAGE, reacts with protein C in non-reduced form only. Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Protein C isolated from human plasma.
Form
Supplied as a liquid in PBS, pH 7.4, 15mM sodium azide.
Purity
Purified by Protein A affinity chromatography from culture supernatant.
Specificity
Recognizes human Protein C. Epitope specificity differs from that of clone 6F1823.
