As major neutral proteases present in the mast cells, tryptases function as mediators in the pathogenesis of asthema and other allergic and inflammatory disorders. Tryptase, beta2 (TPSB2) consists of a signal peptide (residues 1 to 21), a pro peptide (residues 22 to 31) and a mature chain (residues 32 to 275). The amino acid sequence of mouse TPSB2 is 88%, 78%, 76%, and 73% identical to that of mouse, human (TPSB2 and TPSAB1), chimpanzee, and canine.
Applications
Suitable for use in Immunoprecipitation, ELISA and Neutralization. Other applications not tested.
Recommended Dilutions
Immunoprecipitation: 25ug/ml immunoprecipitates recombinant mouse Tryptase, beta 2 from conditioned cell culture medium. Neutralization: Measured by its ability to neutralize Recombinant Mouse Mast Cell Protease6/Mcpt6 (1ug/ml) cleavage of the fluorogenic peptide substrate Mca-RPKPVE-Nval-WRK(Dnp)-NH2 (10uM). The Neutralization Dose (ND50) is typically 3ug/ml. Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recombinant protein corresponding to Ile32-Ser276 from mouse Tryptase, beta2 expressed in NS0 cells.
