Granzymes are neutral serine proteases, which are stored in specialized lytic granules of cytotoxic T lymphocytes (CTLs) and in natural killer (NK) cells. A number of granzymes A to G, have been isolated and cloned from mouse CTLs and NK cells, however in man, fewer have been cloned and identified. One of these, human granzyme B, is involved in target cell apoptosis during lymphocyte mediated cytotoxicity. Exocytosis of granzyme containing granules in the cytoplasm of the target cell leads to the induction of DNA fragmentation and apoptosis. Granzyme B is important in the induction of apoptosis in target cells by cytolytic lymphocytes (CTLs).Granzyme B plays a key role in the induction of apoptosis by CTLs. After delivery to the target cell, Granzyme B activates the cascade of caspases that finally results in cell death.

Applications
Suitable for use in Flow Cytometry, Immunohistochemistry, Immunoprecipitation, Immunofluorescence and ELISA. Other applications have not been tested.
Recommended Dilutions
Flow Cytometry: 1:10-1:100. Use 10ul to label 10e6 cells or 100ul whole blood. Membrane permeabilization is required. ELISA: 2-5ug/ml Immunohistochemistry: Frozen sections Optimal working dilutions to be determined by researcher.
Hybridoma
Spleen cells from immunized Balb/c mice were fused with cells of the mouse SP2/0 myeloma cell line.

Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
