MaxLight™650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor™647, DyLight™649, Cy5™ and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (655nm); Emission (676nm); Extinction Coefficient 250,000.

Expression of p21WAF1 is induced by wild type, but not mutant, p53 suppressor protein. The p21WAF1 protein binds to cyclin/CDK complexes and inhibits their kinase activity thereby stopping cell cycle progression. Meanwhile, the p21WAF1 protein also binds to PCNA (proliferating cell nuclear antigen) and blocks DNA replication but not the DNA repair process. Evidently, inactivation of p53 disrupts the p53-dependent expression of p21WAF1. p21WAF1is a tumor suppressor involved in the pathogenesis of a variety of malignancies.
Pooled monoclonal mouse IgGs
Application(s): Suitable for use in Immunoprecipitation, Western Blot.
Immunoprecipitation: 4ug immunoprecipitates Cip1 from 500ug of HeLa nuclear extract.
Immunoblot Analysis: 0.5–2ug/ml detects WAF1, p21 in 20ug of HeLa nuclear extract. HeLa nuclear extract was resolved by SDS PAGE, transferred to nitrocellulose, and probed with anti-WAF1, p21 (0.5ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system.
Positive Control: HeLa nuclear extract. Use 20ug per lane for minigels.
Storage and Stability
Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™650 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
