MaxLight™ 650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor™647, DyLight™649, Cy5™ and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (655nm); Emission (676nm); Extinction Coefficient 250,000.

Ankyrin G is found in several tissues including brain where, due to tissue-specific RNA processing, it is expressed as two different isoforms, localized to axon initial segments and nodes of Ranvier. Ankyrin G is also found at sites of epithelial cell- cell contact and in the intracellular membranes of some cells. Ankyrin G isoforms contain a unique stretch of sequence highly enriched in serine and threonine residues immediately following a globular head domain. Ankyrin G is thought to participate in the maintenance and targeting of ion channels and cell adhesion molecules to the nodes of Ranvier and to axon initial segments.
Applications
Suitable for use in Immunofluorescence, FLISA, Western Blot and Immunoprecipitation. Other applications not tested.
Recommended Dilutions
FLISA (Native): 0.1-1.0ug/ml Western Blot: 1-3ug/ml. For best results on rat brain, mouse brain or MDCK cell lysates, add 0.3M sucrose to incubation buffer. Detects a band between ~200-300kD. Immunofluorescence (Frozen): 5-10ug/ml. Sensitive to fixation conditions. Recommend 1% paraformaldehde for perfusion and 4 hours of post fixation, after perfusion. For sciatic nerve, recommend 2% paraformaldehyde for 2 hours at 4ºC. Immunoprecipitation (Native): 5ug/ IP Optimal dilutions to be determined by the researcher.
Storage and Stability
Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™650 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
