SETD2 is a histone methyltransferase that exhibits methylation activity toward histones H3-K36.

Intended Use
The SETD2 Direct Activity Chemiluminescent BioAssay™ Kit is designed to measure SETD2 activity for screening and profiling applications.
Test Principle
The key to the SETD2 Chemiluminescent BioAssay™ Kit is a highly specific antibody that recognizes methylated K36 residue of Histone H3. With this kit, only three simple steps on a microtiter plate are required for methyltransferase detection. First, S-adenosylmethionine is incubated with a sample containing assay buffer and methyltransferase enzyme for one hour. Next, primary antibody is added. Finally, the plate is treated with an HRP-labeled secondary antibody followed by addition of the HRP substrate to produce chemiluminescence that can then be measured using a chemiluminescence reader.
Contraindications
Avoid green and blue dyes that absorb light in the Alphascreen® signal emission range (520-620nm), such as Trypan Blue. Avoid the use of the potent singlet oxygen quenchers such as sodium azide (NaN3) or metal ions (Fe2+, Fe3+, Cu2+, Zn2+ and Ni2+). The presence of >1% RPMI 1640 culture medium leads to a signal reduction due to the presence of excess biotin and iron in this medium. MEM, which lacks these components, does not affect Alphascreen® assays.
Applications
Great for studying enzyme kinetics, screening small molecular inhibitors for drug discovery and HTS applications.

Storage and Stability
Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
