This gene encodes an RGD-containing protein that binds to type I, II and IV collagens. The RGD motif is found in many extracellular matrix proteins modulating cell adhesion and serves as a ligand recognition sequence for several integrins. This protein plays a role in cell-collagen interactions and may be involved in endochondrial bone formation in cartilage. The protein is induced by transforming growth factor-beta and acts to inhibit cell adhesion. Mutations in this gene are associated with multiple types of corneal dystrophy.

Intended Use
For the quantitative determination of human transforming growth factor-beta-induced protein ig-h3 (TGFBI/BIGH3) concentrations in serum, plasma and tissue homogenates.
Specificity
This assay has high sensitivity and excellent specificity for detection of human TGFBI. No significant cross-reactivity or interference between human TGFBI and analogues was observed.
Test Principle
This BioAssay™ employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for TGFBI has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TGFBI present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TGFBI is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TGFBI bound in the initial step. The color development is stopped and the intensity of the color is measured.

Storage and Stability
Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
