Mueller Hinton Broth is a general purpose medium that may be used in the cultivation of a wide variety of fastidious and non-fastidious microorganisms. It may also be used for the determination of antibiotic susceptibility by broth dilution method.

Directions per Liter
Dissolve 21 grams in 800-900ml of ddH2O stirring gently with heat. Boil for 1 minute to dissolve the powder completely solubilized. Adjust pH of the medium to the desired level. Add additional water to bring the solution to 1L. Dispense into appropriate containers, loosen caps and autoclave for 15 minutes at 121ºC (15psi).
Note: This medium may be supplemented as required to meet performance criteria. For testing Streptococci, supplementation with 5% defibrinated sheep or horse blood is recommended. Mueller Hinton medium should be supplemented with 2% NaCl for testing methicillin or oxacillin against Staphylococci. Mueller Hinton medium with rabbit serum is used for testing the susceptibility of microorganisms to sulfonamides and trimethoprim.
Storage and Stability
Store powdered medium at 4ºC. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared medium should be kept at 4°C and used within a short period of time.
