Directions per Liter for 100X
Dissolve 3.06 grams in 800-900ml of ddH2O stirring gently with heating until completely solubilized. Adjust pH of the medium to the desired level. Add additional water to bring the solution to1L.
Directions for 500ml S-basal media
2.9g sodium chloride 25ml 1M KH2PO4 475ml DDW 0.5ml cholesterol prepared at 5mg/ml
3. Add the following
5.0ml trace metals solution 1.5ml 1M magnesium sulfate 3.0ml 0.5M calcium chloride. 2. Autoclave. Keep dark (wrap in aluminum foil). 5ml 1M potassium citrate, pH 6.0 5ml100X Pen/Strep/Neo 5ml Nystatin.
Storage and Stability
Store powdered media at RT. Stable for 12 months after receipt. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept at 4°C and used within a short period of time.
