Used to prepare Thermophilus Growth Media
Castenholz TYE media is designed to support the growth of Thermophilus as it is too nutrient-poor to support many other bacterial life.

Directions per Liter Thermophilus Vitamin-Mineral Stock 100X (Powder), T4800
Dissolve 4.052g per liter of ddH2O, stirring gently until completely solubilized. Filter-sterilize using a 0.22 micron membrane filter. Aliquot into sterile containers. Do not autoclave. Contains heat-labile compounds that can be damaged with autoclaving.
Note: The 100X liquid medium forms precipitates at pH values greater than 5. If higher pH is required, it is advisable to prepare a 2X solution from the stock 100X solution and adjust this solution to the desired pH. Filter-sterilize using a 0.22 micron membrane filter. Adjust the volume accordingly for use with Thermophilus Growth Medium Base Broth.
Storage and Stability
Store powdered media at 4°C. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept at 4°C and used within a short period of time.

Directions per Liter Thermophilus Growth Medium Base Broth
Dissolve 9 grams of Thermophilus Growth Medium Base per liter of distilled/deionized (DDI) water. Heat with stirring until completely solubilized. Dispense into appropriate containers. Loosen caps or cover with foil. Autoclave for 15 minutes at 121ºC (15psi). After cooling, asceptically, add 10ml of prepared, sterile Thermophilus Vitamin-Mineral Stock 100X (T4800) to sterile Thermophilus Growth Medium Base Broth. |
