Cytokine that plays an essential role in the regulation of survival, proliferation and differentiation of hematopoietic precursor cells, especially mononuclear phagocytes, such as macrophages and monocytes. Promotes the release of proinflammatory chemokines, and thereby plays an important role in innate immunity and in inflammatory processes. Plays an important role in the regulation of osteoclast proliferation and differentiation, the regulation of bone resorption, and is required for normal bone development. Required for normal male and female fertility. Promotes reorganization of the actin cytoskeleton, regulates formation of membrane ruffles, cell adhesion and cell migration. Plays a role in lipoprotein clearance.

Applications
Suitable for use in FLISA, Western Blot, Immunohistochemistry and Neutralization. Other applications not tested.
Recommended Dilution
FLISA: To detect hM-CSF by sandwich FLISA (using 100ul/well antibody solution) a concentration of 0.5-2ug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotinylated Anti-Human M-CSF as a detection antibody, allows the detection of at least 0.2-0.4ng/well of recombinant hM-CSF. Western Blot: To detect hM-CSF by Western Blot analysis this antibody can be used at a concentration of 0.1- 0.2ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hM-CSF is 1.5-3ng/lane, under either reducing or non-reducing conditions. Immunohistochemistry: This antibody stained formalin-fixed, paraffin-embedded sections of human cervical squamous cell carcinoma. The recommended concentration is 0.5ug/ml with an overnight incubation at 4°C. An HRP-labeled polymer detection system was used with a DAB chromogen. Heat induced antigen retrieval with a pH 6.0 sodium citrate buffer is recommended. Neutralization: To yield one-half maximal inhibition of the biological activity of hM-CSF (7ng/ml), a concentration of 0.02-0.03ug/ml of this antibody is required. Optimal dilutions to be determined by the researcher.
Storage and Stability
Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Note: Applications are based on unconjugated antibody.
